RESUMEN

Glioblastoma (GBM) is the most common malignant primary brain tumor. Despite comprehensive treatment with traditional surgery, radiotherapy, and chemotherapy, the median survival rate is <14.6% and the 5-year survival rate is only 5%. FBXO22, a substrate receptor of the SCF ubiquitin ligases, has been reported to play a promoting role in melanoma, liver cancer, cervical cancer, and other cancers. However, the function of FBXO22 in GBM has not been reported. In the present study, we demonstrate that FBXO22 is highly expressed in glioma and is positively correlated with worse pathological features and shorter survival of GBM patients. We revealed that FBXO22 promotes GBM cell proliferation, angiogenesis, migration, and tumorigenesis in vitro and in vivo. In terms of mechanism, we reveal that FBXO22 decreases VHL expression by directly mediating VHL ubiquitination degradation, which ultimately increases HIF-1α and VEGFA expression. In addition, our data confirm that there are positive correlations among FBXO22, HIF-1α, and VEGFA expression, and there is a negative correlation between FBXO22 and VHL protein expression in glioma patients. Our study strongly indicates that FBXO22 is a promising diagnostic marker and therapeutic target for glioma patients.

In recent years, the poultry industry had been markedly affected by adenoviral diseases such as hydropericardium syndrome and inclusion body hepatitis caused by fowl adenovirus (FAdV), which have become increasingly prevalent in China. Shandong Province, China, is an important area for poultry breeding where various complex and diverse FAdV serotypes were isolated. However, the dominant strains and their pathogenic characteristics are not yet reported. Therefore, a pathogenicity and epidemiological survey of FAdV was conducted, showing that the local dominant serotypes of FAdV epidemics were FAdV-2, FAdV-4, FAdV-8b, and FAdV-11. Their mortality rates in the 17-day-old specific-pathogen-free (SPF) chicks ranged from 10 to 80%; clinical signs included mental depression, diarrhea, and wasting. The maximum duration of viral shedding was 14 days. The highest incidence in all infected groups was on days 5-9, and then gradual regression occurred thereafter. The most pronounced symptoms occurred in chicks infected with FAdV-4, including pericardial effusion and inclusion body hepatitis lesions. Our results add to the current epidemiological data on FAdV in poultry flocks in Shandong and elucidate the pathogenicity of dominant serotypes. This information may be important for FAdV vaccine development and comprehensive epidemic prevention and control.

TEMPO oxidized nano-fibrillated cellulose (TONFC) has been used in different applications including biomedical, packing materials, paints and cosmetics because of its higher transparency, mechanical properties and better biocompatibility. However, pulping is always required to remove lignin and hemicellulose, and high-energy homogenization is required to defibrillate cellulose bundle into filament. Therefore, it is desirable to find a novel way to get TONFC with high carboxyl content without intensive mechanical disintegration. In this work, nanocellulose (TOHOLO) with higher carboxyl groups (2.2 ± 0.2 mmol/g) and smaller size (length = 400-685 nm and diameter = 5.9 nm) was prepared by a two-step strategy without intensive mechanical homogenization. In addition to the advantages in terms of diameter and carboxyl groups, TOHOLO showed better transparency and re-dispersibility as well as higher mechanical properties (122.8 MPa) compared to previous reports. Furthermore, for high carboxyl group and dispersibility, the TOHOLO can be used as a reinforcing filler to fabricate nanocomposites. The reinforced PVA fibers show a tensile strength of 484.4 MPa, which is about 170 % higher than comparison samples (PVA/TOCN-M, 291.9 MPa).

The study of the impact of high temperature and drought on the yield of major staple crops can provide important scientific support for the decision-making of agricultural sustainable development. Based on the temperature and precipitation data of the European Centre for Medium-Range Weather Forecasts (ECMWF) ERA 5 for northern China, this paper calculates three indexes, the standard precipitation index (SPI), standardized precipitation evapotranspiration index (SPEI) and the extreme degree-day (EDD), from 1979 to 2017. Monthly SPI and monthly SPEI were calculated at 1 - to - 12 month lags, and EDD was calculated per crop growth season. The yield of winter wheat, spring wheat and summer maize in each province of the study area from 1979 to 2017 was de-trended, and the relative fluctuation of the yield of the three crops was calculated. The change trends of SPI, SPEI and EDD were analysed using the Mann-Kendall test and Sen's slope. The single and interactive effects of high temperature and drought on crop yield were studied using multidimensional Copula function. The results show that: 1) Both high temperature and drought stress in northern China show an increasing trend. The drought trend in the study area detected based on SPEI was greater than the drought trend detected by SPI. The difference between SPEI and SPI in the winter wheat growing season was smaller than that in the spring wheat and maize growing seasons. 2) With the increase in EDD and the decrease of SPI/SPEI values, the probability of negative yield fluctuation gradually increased, and the probability of positive yield fluctuation gradually decreased. Under the same drought and high temperature conditions, the probability of yield fluctuation varies among different crops and different provinces. Drought has a greater impact on crop yield than high temperature. Both the single and interactive effects of drought and high temperature on yield are nonlinear. 3) Irrigation can effectively alleviate the impact of drought and high temperature on yield. In heavily irrigated provinces, the effects of both high temperature and drought on crop yield are not obvious.

The present study aims to investigate the effects of Xuesaitong (XST)injection on inflammation induced by OGD/reoxygenation in BV2 microglia cell and explore the underlying mechanisms.The effects of XST injection were evaluated in terms of cell viability, secretion of TNF-α, IL-1ß, IL-6 and IL-10 into culture media, protein expression of p-ERK1/2, p-JNK and p-p38, and nuclear translocation of NF-κB p65. The results showed that XST injection significantly increased cell viability, suppressed release of TNF-α, IL-1ß, IL-6 and IL-10 and down-regulated p-JNK1/2 and p-p38 MAPK expression in BV2 microglia cells induced by OGD/R injury, whereas it had no effect on p-ERK1/2 expression. Furthermore, XST injection suppressed nuclear translocation of NF-κB p65 in BV2 microglia after OGD/R injury. These data indicate that the neuroprotective effects of XST injection on OGD/R injury are associated with its inhibition of pro-inflammatory mediator production, down-regulation of JNK1/2 and p38 MAPK activation, and suppression of NF-κB p65 nuclear translocation in BV2 microglia cells.

E2F3a, as a member of the E2F family, is essential for cell division associated with the progression of many cancers. However, the biological effect of E2F3a on glioma is not understood as well. To investigate the functional mechanism of E2F3a in glioma, we examined the expression of E2F3a in glioma tissue and cell lines. We found that E2F3a was upregulated in glioma tissue compared with adjacent tissue, and this was associated with a poor survival rate. E2F3a was highly expressed in glioma cell lines compared with normal HEB cell lines. Knockdown of E2F3a significantly inhibited cell proliferation, promoted G0/G1 phase arrest, elevated apoptosis rates, and suppressed cell migration and invasion. However, overexpression of E2F3a markedly promoted cell proliferation, migration, and invasion and inhibited apoptosis. Moreover, in vivo studies showed that knockdown of E2F3a expression dramatically inhibited U373 tumor growth in a nude mouse model. Results of real-time PCR and Western blot showed that the depletion of E2F3a upregulated the expression levels of cell apoptosis-related proteins and downregulated migration-related proteins. Conversely, E2F3a overexpression downregulated the expression levels of cell apoptosis-related proteins and upregulated migration-related proteins. In conclusion, our results highlight the importance of E2F3a in glioma and provide new insights into the diagnostics and therapeutics of gliomas.

RESUMEN

We propose and experimentally demonstrate switchable operation of dissipative solitons (DSs) in an erbium-doped fiber laser. By managing normal dispersion of laser cavity, the 3-dB spectral bandwidth up to 8.1 nm can be obtained with the help of a semiconductor saturable absorber mirror. Using an inline polarizer, the fiber laser can be separately operated at either wavelength-tunable or multi-wavelength emission. The central wavelength of DS can be continuously tuned from 1554 to 1561 nm with its spectra maintaining standard rectangular shape. Alternatively, triple-wavelength DSs at 1535, 1544, and 1553 nm can be simultaneously obtained.
